چکیده

I n tr o d u c tio n T E cfi'ccls o f the systemic adminis tration o f crude papain to young rabbits were first described by Thomas (1956), who noted that the most obvious change was a bilateral collapse o f the ears. This effect was lost within a few days. Subsequently llu lth and Westerborn (1959) and Merkow and Lalieh (1961), who iLsecl crude papain in single and repeated (loses, and in different species o f animals, not only confirmed but also elaborated on the effects o f its administration. After finding that crystalline papain failed to produce collapse o f rabbit ears, even when injected in large amounts, McClusky and Thomas (1955!) observed that i f this enzyme preparation was in activated before administration by the addition o f a thiol-combining agent such as iodo-acctamide, the typical collaji.se did occur. Th e inactivated enzyme, how ever, did not produce this effect in vitro. This study was undertaken to deter mine the effects o f local and systemic in jections o f inactivated crystalline papain in rabbits and rats, with a view to its possible clinical application in the early treatment o f bilateral cleft palates.

Thermoanalytical and histological study of intra-articular papain-induced degradation and repair of rabbit cartilage. II. Mature animals.

The effect of papain treatment on the cartilage tissue of the knee joint of mature rabbits was studied by histological and thermoanalytical methods. Changes taking place in the composition of cartilage in the course of ageing were shown. As a result of papain treatment a significant irreversible destruction of the ground substance took place. Papain-induced loss of proteoglycans and the intensi...

The Removal of Cartilage Matrix, in Vivo, by Papain

The intravenous injection of crystalline papain into young rabbits results in depletion of cartilage matrix throughout the body, with loss of rigidity and collapse of the ears, provided the enzyme is inactivated by oxidation or sulfhydryl blocking agents prior to administration. Cysteine-activated crystalline papain, when injected intravenously, produces little or no change in cartilage. The ch...

Increased physical activity severely induces osteoarthritic changes in knee joints with papain induced sulfate-glycosaminoglycan depleted cartilage

INTRODUCTION Articular cartilage needs sulfated-glycosaminoglycans (sGAGs) to withstand high pressures while mechanically loaded. Chondrocyte sGAG synthesis is regulated by exposure to compressive forces. Moderate physical exercise is known to improve cartilage sGAG content and might protect against osteoarthritis (OA). This study investigated whether rat knee joints with sGAG depleted articula...

Papain-induced Changes in Rabbit Cartilage

Some biochemical aspects of the collapse of the rabbit ears produced by the intravenous injection of papain have been studied. A marked depletion of chondromucoprotein (M.C.S.) and a reduction of the S(35) content of cartilage matrix were found to coincide with the gross and histologic changes in the cartilage. At the same time there was a marked increase in the amount of S(35) in the serum and...

Warm sparse-dense wave inhibits cartilage degradation in papain-induced osteoarthritis through the mitogen-activated protein kinase signaling pathway

Cartilage degradation is an important in the pathogenesis of osteoarthritis (OA). Abnormal activation of the mitogen-activated protein kinase (MAPK) signaling pathway in chondrocytes promotes an inflammatory response, resulting in the release of chondral matrix-degrading enzymes that accelerate the degradation of cartilage. As a non-pharmaceutical and non-invasive physical therapy regimen, warm...

Comparison of the Effects of Papain and Vitamin a on Cartilage

The effects of papain protease and of vitamin A on explanted limb bone rudiments from 7- and 13-day chick embryos and fetal mice have been studied and compared. The incubation of cartilaginous rudiments from 7-day chick embryos in a solution containing papain and cysteine resulted in complete loss of the metachromasia of the cartilage matrix within 1 hour; explants treated in this fashion recov...
